If you are referring to act 2 they know them from the previous day. And they do not pretend but rather they don't remember. Vladimir has an idea of who they are but is not completely certain due to Estragon not remembering. This show the tragedy in loss of memory. Throughout the whole play the two characters have many episodes where they forget. They even forget each other, on occasion. In act 1 they just met him. And they aren't exactly "pretending" they are just extremely clownish. Which is how they find it best to pass the time and wait.
